DL-Phenylalanine is a composite Nootropic amino acid comprised of two stereoisomers (D- and L-) of the amino-acid phenylalanine. L-Phenylalanine occurs naturally in meats and protein-rich foods and is responsible for a number of effects including mood-lift, stress-reduction and an increase in energy and focus. It is an essential amino acid and is required for a large variety of functions in the body and is a precursor to tyrosine (important in producing epinephrine and norepinephrine).

D-Phenylalanine is lab synthesized and is responsible for blocking pain signals. DL-Phenylalanine is a Nootropic with the combined effects of the two isomers and is important in boosting levels of norepinephrine and dopamine in the brain – thereby improving focus, increasing mood and energy and reducing sensations of pain. DL-Phenylalanine benefits / effects


DL-Phenylalanine appears to have a variety of benefits to its use. These benefits include effects such as: mood-lift, increased energy and mental focus and pain relief. DL-Phenylalanine has also been researched as a good support for healthy mood, potentially helping to reduce chronic pain and also for increasing mental clarity.

L-Phenylalanine is known to help the brain to produce neurotransmitters like epinephrine, norepinephrine and dopamine. These are crucial to proper functioning of the brain and the first two result in an increase in energy and focus, while dopamine is known to lift the mood and reduce stress and anxiety.

D-Phenylalanine is known to decrease functioning of the enkephalinase enzyme, which may result in pain relief. Also, taking DL-Phenylalanine may lower the side effect of raised blood pressure associated with taking L-Phenylalanine alone.

There is still room for a lot more research into the mechanisms, benefits and actions of both D- and L-Phenylalanine and specifically D-Phenylalanine which is synthesized in laboratories and not found in nature.[2]


Research into treatment of depression

There have been many studies into the effects of DL-Phenylalanine for the treatment of depression. In one such study, 20 people with diagnosed clinical depression were given 75-200mg DL-phenylalanine over a course of 20 days. At the end of the study, 60% of the patients had no symptoms of depression, and 20% were able to be discharged from hospital.

A separate, individualised trial using L-Phenylalanine in combination with MAO inhibitors on 10 patients with drug resistant major depression found that after three days of treatment, nine of the ten patients had found considerable mood-elevation hours after dosing, and six of the ten considered their conditions to have been fully treated.

Two separate double-blind studies using doses of 100mg D-Phenylalanine or 150mg DL-Phenylalanine found that, over 30 days of treatment the majority of patients had improved psychologically and clinically in their conditions[3].


Research into treatment of chronic pain

D-Phenylalanine is thought to prevent the action of the enkephalinase enzyme, which is responsible for the break-down of enkephalins – molecules that reduce the sensation of pain. This is thought to be the mechanism by which D-Phenylalanine is able to produce pain-relief and possibly act as a treatment for chronic pain.  This is also known as up-regulation of the ‘endogenous analgesia system’ (EAS).[4]


Other possibilities for clinical treatments

As mentioned above, there is room for much more research into the possibilities of DL-Phenylalanine based treatments. Some preliminary research has shown that DL-Phenylalanine may be helpful in the treatment of Parkinson’s Disease but there is a need for much more research into this.

Phenylalanine in various forms has been offered as a possible treatment for the currently untreatable skin condition known as vitiligo, where white, pigment-free patches develop all over the skin. This also requires far more research and testing.

DL-Phenylalanine has also been proposed as a great supplement for improving mental clarity and focus.[5]


As a supplement, or part of a supplement stack

The L-Phenylalanine component of DL-Phenylalanine is used as one of the main, protein forming amino acids in every area and aspect of the body; therefore supplementing with DL-Phenylalanine can help ensure adequate levels of this important amino acid.  DL-Phenylalanine has a number of additional proposed effects and benefits but there is not really enough research out there to indicate whether it is properly effective in these areas when taken on its own. It may be an option to consider taking DL-Phenylalanine in conjunction with other Nootropics or in small doses as a supplement ‘stack’ to gain the most benefits in health.


DL-Phenylalanine recommended use


There is a large amount of information online about the correct dosage of DL-Phenylalanine. It is not absorbed into the blood stream as efficiently as L-Phenylalanine or D-Phenylalanine alone, so doses are normally a little higher than when taking either of the stereoisomers alone.

Recommended serving sizes appears to be from around 100 - 500mg daily although some websites do suggest that users may benefit from taking up to 1000mg a day.


DL-Phenylalanine side effects and warnings


There have been some reported side effects of DL-Phenylalanine (also known as DLPA) use. These side effects may include heartburn, nausea and headaches as well as increased blood pressure. L-Phenylalanine alone is known to increase blood pressure considerably which is why using DLPA may be a better option.

L-Phenylalanine has been used alone in supplements for a while, but is known to increase blood pressure. DL-Phenylalanine does not appear to exhibit this effect to such an extent.

At high doses of 1000-1500mg, users have reported feelings of numbness and tingling sensations. Prolonged use of DL-phenylalanine at high doses may cause nerve-damage.

DL-Phenylalanine should not be taken by people with the rare disorder known as phenylketonuria (PKU).[6]
